Protocols
Full plasmid sequencing service module

Pooled plasmid library preparation and verification. This workflow enables the recovery, expansion, and sequence verification of pooled plasmid libraries. Plasmids are first rescued from yeast colonies through DNA extraction and then introduced into E. coli by high-efficiency transformation. Individual clones are randomly picked, arrayed, and replicated before being pooled for transposome-based library preparation. The resulting libraries are then sequenced to verify and index the assembled DNA blocks.
TAR assembly service module

Yeast-based assembly of synthetic DNA segments. Synthetic DNA segments are divided into several overlapping fragments with unique homology regions, including terminal homology to an acceptor plasmid. The fragments and acceptor plasmid are introduced into yeast, where homologous recombination assembles them into a complete DNA segment within the plasmid.
